Mechanistically, cellular senescence causes cells to permanently exit the cell cycle while remaining metabolically active and secreting pro-inflammatory signals (the senescence-associated secretory phenotype, or SASP). Because this interaction shields senescent cells from programmed cell death, FOXO4-DRI functions as a selective senolytic agent that restores p53-mediated apoptosis without damaging healthy, non-senescent cells.
Consequently, preclinical studies have examined whether the targeted elimination of senescent cells via FOXO4-DRI can restore tissue homeostasis, suppress SASP-driven systemic inflammation, and improve functional outcomes in aging models. In addition, research has demonstrated positive effects on physical endurance markers, fur density recovery, and renal function in aged animals. Furthermore, its retro-inverso structural design protects the peptide against proteolytic degradation, significantly extending its stability in vivo. Overall, FOXO4-DRI represents one of the most precise molecular senolytic tools available for investigating the causal role of cellular senescence in geroscience.
